内容:The 6-kDa early secreted antigenic target (ESAT-6) is an essential virulence factor for pathogenesis of Mycobacterium tuberculosis (Mtb). Through its membrane-interacting activity, ESAT-6 mediates translocation of Mtb from the phagosomes to the cytosol within host macrophages, a critical step in the transition of a latent infection to active tuberculosis (TB) diseases. Thus, ESAT-6 is an important therapeutic target for inhibiting the transition from latent to active TB. ESAT-6 is also a potent T-cell antigen and uniquely present in virulent mycobacterial species, but not in avirulent species, such as TB vaccine strains, which makes ESAT-6 an excellent candidate for development of novel vaccines and diagnosis against TB. The ESAT-6-centered research in Dr. Sun’s laboratory is integrating multidisciplinary approaches to investigate the mechanism of ESAT-6 membrane interaction and develop novel vaccines and diagnostic approaches to fight the global TB epidemics.
